Some genes such as ISZ-sTRAIL come from the NCBI website, and then primers are designed according to the plasmids sent by the company, and the target genes are first obtained by PCR. After that, the obtained target gene and pET28a empty load are used for double digestion with Nco 1 and Xho 1; after digestion, the digested product is ligated and transformed. Finally, the plasmid is extracted from the transformed bacteria for double enzyme digestion verification, and the verified plasmid is sent for sequencing.
